A mushroom plate culture is a 100mm agar petri dish that has been pre-colonized with live, actively-growing mushroom mycelium of a specific species. You can see the mycelium clearly on the plate before using it, which lets you verify genetics and contamination before transferring. Plate cultures are the preferred starting point for serious cultivators building a culture library.

Both contain live mycelium, but a plate culture grows on solid agar media in a petri dish while a liquid culture grows in sterile nutrient broth inside a syringe. Plate cultures let you visually inspect for contamination and produce stronger, more vigorous genetics. Liquid cultures are faster for direct injection into substrate. Most cultivators use plates to maintain their genetic library and liquid culture for bulk inoculation.

Work in front of a laminar flow hood or inside a still air box. Sanitize your scalpel or agar punch with isopropyl alcohol. Cut a small wedge of clean, white mycelium (avoid the edges). Drop the wedge directly into a sterilized grain bag through the injection port, or into a liquid culture broth jar. One plate can typically inoculate 2–4 grain bags or multiple liquid culture jars.

One plate culture can typically be expanded into 8–12 new plates through wedge transfers. This is how cultivators build and maintain a genetic library — one $26 plate becomes the foundation for dozens of plates over multiple generations. For best results, transfer from young, actively-growing mycelium rather than older plates.

Refrigerate the plate culture upon receipt. Mycelium goes dormant at refrigerator temperatures, extending shelf life for weeks. Use as soon as possible for the healthiest, most vigorous mycelium. Do not freeze — this will crack the agar and kill the culture.

A laminar flow hood is highly recommended for plate work because agar is an open medium that is very susceptible to airborne contamination. A still air box (SAB) can work for careful beginners, but a flow hood dramatically improves your success rate. For transfers into sealed substrates like grain bags with injection ports, a still air box is often sufficient.

Spore syringes contain dormant, genetically-mixed spores that can take weeks to germinate and produce inconsistent results. A plate culture contains live, vigorous mycelium from a lab-selected isolate — proven genetics, faster colonization, and uniform fruiting behavior. Plate cultures also let you verify contamination visually before using them, which spore syringes cannot.
