How to Use Pre-Poured Agar Plates

Whether you are germinating spores, cloning tissue, or transferring mycelium โ€” the process is simple.

Prepare

Work in front of a laminar flow hood or inside a still air box. Wipe all surfaces and tools with isopropyl alcohol.

Inoculate

Remove the Parafilm seal. Drop spore solution, place a tissue clone, or transfer a wedge of mycelium from another plate onto the agar surface.

Seal & Incubate

Re-seal the plate with Parafilm or lab tape. Incubate in a clean environment at approximately 76ยฐF. Growth appears within 3โ€“10 days depending on species and inoculation method.

Transfer or Use

Once colonized, transfer clean sectors to fresh plates for isolation, or use the culture to inoculate liquid culture, grain spawn, or BRF jars.
